More questions about achieving Heterochromia iridum

Hey again
So... In my last post about this I asked if it was possible to achieve Heterochromia iridum(A mutation that causes your eyes to have different colors) through energy programming. Thanks to exaarkun I now know that it is indeed possible, but an advanced working. I would like to know how exactly such a working should be done. If you raise the energy and program it with for example "My left eye is safely and positively changing its color to...", should you do this together with a specific rune? Is there a specific amount of repititions for things like these? And where should the energy be directed to? To the eye or to the aura?
I'm very curious, I know. But it fascinates me that something like this is possible.

I might do this as a side-thing if I find the time, so no it's not my main goal, I'm just pretty curious about it. 
And thanks, I will say "Iris" instead. 
Any thoughts on the repititions/runes and where I should direct the energy?
